El cerebelo es una parte del cerebro que ayuda a coordinar y regular una amplia gama de funciones y procesos tanto en el cerebro como en el cuerpo. La degeneraci\u00f3n del cerebelo en la AT causa graves problemas de movimiento y hace que los ni\u00f1os se conviertan en usuarios de silla de ruedas a tiempo completo alrededor de los 10 a\u00f1os. Actualmente no existe una terapia cl\u00ednicamente aprobada para tratar la degeneraci\u00f3n cerebelosa inducida por la AT. Por lo tanto, el objetivo de este proyecto es desarrollar un enfoque terap\u00e9utico que trate la causa ra\u00edz de esta degeneraci\u00f3n.<\/p><p class=\"p1\">No existe ninguna terapia cl\u00ednicamente aprobada que restaure la expresi\u00f3n de ATM en el cerebelo. ATM es la prote\u00edna que no funciona correctamente o que falta en ni\u00f1os y adultos con AT. Algunas c\u00e9lulas que se encuentran en el cerebelo no funcionan bien sin ATM y mueren gradualmente, lo que causa degeneraci\u00f3n.\u00a0<\/p><p class=\"p1\">La terapia g\u00e9nica es una forma de tratar o prevenir una enfermedad espec\u00edfica mediante la introducci\u00f3n de genes que funcionan normalmente en las c\u00e9lulas en lugar de los que faltan o son defectuosos para corregir trastornos gen\u00e9ticos. Una forma com\u00fan que los investigadores han descubierto para lograr esto es mediante el uso de un vector. Los vectores son esencialmente veh\u00edculos dise\u00f1ados para administrar material gen\u00e9tico terap\u00e9utico, como un gen funcional, directamente a una c\u00e9lula.<\/p><p class=\"p1\">Los investigadores utilizar\u00e1n vectores adenovirales dependientes de ayudantes (HdAd). Estos vectores tienen perfiles de seguridad mejorados y una gran capacidad para transportar material gen\u00e9tico. HdAd proporciona expresi\u00f3n g\u00e9nica a largo plazo sin toxicidad despu\u00e9s de una sola administraci\u00f3n en m\u00faltiples modelos animales precl\u00ednicos. Debido a estas caracter\u00edsticas, HdAd es extraordinariamente prometedor para tratar la AT. Actualmente, su uso en el cerebelo no se ha explorado ya que los vectores HdAd actuales no funcionan para las c\u00e9lulas de Purkinje (neuronas grandes que se encuentran en el cerebelo y que se ven afectadas en las personas con AT). Por lo tanto, existe una brecha de conocimiento en el desarrollo de enfoques de terapia g\u00e9nica HdAd para tratar la degeneraci\u00f3n cerebelosa por AT.<\/p><p class=\"p1\">Bas\u00e1ndose en la d\u00e9cada de experiencia del laboratorio en el desarrollo y uso de vectores HdAd en el sistema nervioso central, el equipo ha creado un nuevo vector HdAd que transduce c\u00e9lulas de Purkinje y otros tipos de c\u00e9lulas cerebelosas en un modelo de rat\u00f3n. Ahora llevar\u00e1n a cabo trabajos para superar las principales limitaciones que impiden el uso de HdAd en el cerebelo y su objetivo es desarrollar un enfoque HdAd que proporcione niveles terap\u00e9uticos de ATM y caracterice su eficacia en la restauraci\u00f3n de la funci\u00f3n ATM en las c\u00e9lulas de Purkinje. Utilizar\u00e1n sus nuevos vectores virales para desarrollar enfoques para la expresi\u00f3n y restauraci\u00f3n a largo plazo de la funci\u00f3n ATM en las c\u00e9lulas de Purkinje y otros tipos de
