class=\"p1\"><b>Objetivo:<\/b>\u00a0Los medicamentos que probar\u00e1n han sido seleccionados expresamente para que sea m\u00e1s r\u00e1pido y sencillo pasar a las primeras etapas de los ensayos cl\u00ednicos. Todos los medicamentos son antiguos y de uso com\u00fan, lo que significa que son baratos y no tienen problemas de patentes, adem\u00e1s de que sus perfiles de seguridad y posibles efectos secundarios son bien conocidos.<\/p><p class=\"p1\">Es importante destacar que se sabe que todos ellos penetran en el cerebro y varios ya se utilizan para tratar enfermedades neurol\u00f3gicas, incluidas enfermedades que aparecen en la infancia, como la epilepsia. Si pueden ver efectos positivos de uno o m\u00e1s medicamentos en sus c\u00e9lulas, moscas de la fruta y minicerebros, avanzar\u00e1n hasta las siguientes etapas: ensayos cl\u00ednicos para probar si son capaces de ayudar a los pacientes con AT a mantener la funci\u00f3n neurol\u00f3gica durante m\u00e1s tiempo.<\/p><p class=\"p1\">Los pacientes con ataxia telangiectasia (AT) tienen una coordinaci\u00f3n deficiente de movimientos porque las c\u00e9lulas nerviosas de una parte del cerebro comienzan a funcionar mal y finalmente mueren. Se cree que un proceso particular conocido como autofagia es defectuoso en las c\u00e9lulas nerviosas de AT. La autofagia es la forma en que las c\u00e9lulas reciclan materiales y componentes viejos para su reutilizaci\u00f3n. Se sabe que este proceso de reciclaje es defectuoso en casi todas las formas de enfermedad neurol\u00f3gica, incluso en otros trastornos hereditarios raros que afectan al sistema nervioso. Se sabe que muchos medicamentos existentes aumentan los niveles de reciclaje en las c\u00e9lulas. En este proyecto, van a buscar entre esa gama de medicamentos para tratar de identificar uno o m\u00e1s que puedan restaurar el reciclaje en las c\u00e9lulas nerviosas de AT con la esperanza de que estos medicamentos puedan reutilizarse para tratar a los pacientes.<\/p><p class=\"p1\">Proceso:\u00a0<\/p><p class=\"p1\">Primero se cultivar\u00e1n c\u00e9lulas nerviosas de AT en el laboratorio y se probar\u00e1 una selecci\u00f3n de medicamentos que se sabe que mejoran el reciclaje. Si algunos parecen prometedores, los probar\u00e1n en dos modelos m\u00e1s complejos de AT. Primero, utilizar\u00e1n moscas de la fruta que han sido dise\u00f1adas para reflejar lo que sucede en AT. Las moscas de la fruta, aunque mucho m\u00e1s simples que los humanos, tienen un sistema nervioso complejo y muestran un comportamiento complejo. Buscar\u00e1n ver si alguno de los medicamentos corrige los cambios estructurales en los nervios que observan en sus moscas AT que se deben a un reciclaje defectuoso. Tambi\u00e9n probar\u00e1n si los medicamentos son capaces de corregir los d\u00e9ficits de movimiento que muestran las moscas.<\/p><p class=\"p1\">En la \u00faltima fase del proyecto, se crear\u00e1n minicerebros en el laboratorio. A lo largo de varias semanas, estos minicerebros, conocidos como organoides, se convertir\u00e1n en estructuras tridimensionales complejas con muchos tipos de c\u00e9lulas diferentes y caracter\u00edsticas que recuerdan a un cerebro. Ya saben que en los organoides cerebrales creados a partir de c\u00e9lulas AT, los genes asociados con el proceso de reciclaje de la autofagia no se activan y desactivan correctamente y que hay un aumento de la muerte de c\u00e9lulas nerviosas. El equipo espera que los medicamentos que han demostrado funcionar en las c\u00e9lulas y en las moscas de la fruta puedan revertir el reciclaje defectuoso en los organoides cerebrales y proteger de la
